No clearance due to no runway assigned?

Having connected PF3 to FSX, and loaded a flightplan in both, when I attempt to request clearance, I invariable see a message xomething like "Unable provide clearance until runway assigned" !!!
My question is, what and when causes PF3 to assign takeoff runway and is able to provide clearance? How long do I need to wait or is there something I should be setting that maybe I'm not?

If you press 6 before you have pressed 0 to say you're ready to taxi then PF3 will give that message.
