I have recently stareted using MSFS with PF3 (FS2020) and cannot get the 'roger' hotkey combination to work/readback. I have installed 'runtimes', tried changing/re-assigning keys but no joy. I have enabled the 'beep on hotkey' press and there is no 'beep' when pressing the combination CTRL+Shift+\. Any ideas ?

If you're not hearing a beep when hitting the hotkey then it simply isn't being recognised so you need to assign another key. Try something simply to start with like using a single key for the roger key. There are also a few posts on the forum about this issue which might be worth checking out.

As Dave said you can try experimenting with a single hotkey. Also check out this thread which describes a similar issue (see the second page of the thread as well).
